* Calculated from the zero-time viable spore concentration taken as 0 per cent germination. The data shown in table 1 indicate that the phenol-inactivation method and the dilution heat-shock method were superior to the conventional heat-shock method usually employed in germination studies if germination rates are desired. This was especially true for short incubation times when the germination of spores was less than 99 per cent. At longer incubation times when germination is greater than 99 per cent, any one of the three methods is adequate. Also, the data in table 1 show a good correlation between the dilution heat-shock technique and the phenol-inactivation method. Phenol treatment to inactivate germinated or vegetative cells of B. anthracis is recommended as the best method for determining the numbers of ungerminated spores in a culture because it does not require a heated water bath, is simple to perform, is instantaneous in destroying heatlabile cells, and correlates well with accurately performed heat-treatment methods when germination or sporulation rates are desired.
